Anti-EphA3 Program (KB004)

Humaneered™ Antibody Targeting EphA3 for the Treatment of Cancer

PRODUCT DESCRIPTION

KB004 is a first-in-class engineered human IgG1 antibody targeting the EphA3 receptor tyrosine kinase. Identified using KaloBios proprietary antibody Humaneering™ technology, KB004 has Variable regions very close to human germ-line sequences providing the potential for reduced immunogenicity in patients.

EphA3: A TARGET FOR TUMOR NEOVASCULATURE PLUS SOLID & HEMATOLOGIC MALIGNANCIES

EphA3 receptor tyrosine kinase is selectively expressed on the surface of multiple solid tumors and hematologic malignancies and also plays an important role in tumor neovasculature. Anti-EphA3 therapy has the potential to kill tumors through several distinct mechanisms:
  • Tumor neovasculature disruption: agonist activity causes shape-change in tumor endothelial cells
  • Tumor cell killing: potent ADCC activity and direct apoptosis
With more than one way to attack tumors, KB004 may have broad applicability to treat most tumor types both as a single agent and in combination with other therapeutics.

PRECLINICAL DATA: PROOF-OF-CONCEPT ESTABLISHED

EphA3 expression has been documented in multiple tumor types including: melanoma, breast, non-small cell lung, colon, renal cell, glioblastoma, prostate, and leukemia. KB004 binds to both mouse and human EphA3 with similar affinity, allowing the effects on tumor neovasculature to be studied in tumor xenograft and syngeneic models in mice. To date, anti-EphA3 has shown encouraging preclinical proof-of-concept results in multiple tumor models. Furthermore, the biodistribution of anti-EphA3 and other initial safety-related data indicate that KB004 may be well-tolerated in humans. Comprehensive preclinical data may be shared confidentially.



FUTURE DEVELOPMENT

KaloBios plans to file an IND for KB004 in Q3 2010, and start clinical testing in hematological malignancies in the United States in Q4.
