Screening
The ‘Epitope Focused’ library is transformed into a microbial host cell and plated onto growth medium. Fab fragments are expressed using a proprietary secretion system from E. coli using a mutant strain that allows secretion of correctly folded and assembled antibody fragments without the need for signal sequences or for surface display. One method of selection from the library is via our Colony Lift Binding Assay. A filter, impregnated with the target, is challenged with the 'Epitope Focused' library. The filters are washed stringently enough that only antibodies of higher affinity than the starting murine antibody will remain. Clones are chosen for subsequent analysis.
