The Colony Lift Binding Assay (shown below) demonstrates how panels of clones,
all with the identical specificity, are produced; however, the composition and
physical characteristics of each clone is distinct, affording ample opportunity
to screen for clones containing all the desired properties of the target
therapeutic.
Clones are selected for Ka, Kd, closeness-to-human germline, stability,
expression, aggregation, etc. Below are shown the aggregation profiles
(SEC-HPLC) for five Humaneered™ clones. Two clones show undetectable aggregation
and will be included for further analysis on the screening cascade.
